Tàu vũ trụ Vostok 3KA chỉ mới được phóng thử nghiệm một lần trước đó, vào ngày 9/3/1961, trong sứ mệnh Korabl-Sputnik 4, sứ mệnh đã diễn ra thành công.[4] Trước khi sứ mệnh Korabl-Sputnik 4 diễn ra, Liên Xô đã thực hiện hai sứ mệnh vào tháng 12 năm 1960, cũng trong khuôn khổ chương trình Vostok nhưng cả hai đều gặp thất bại.[5]

Only days before the launch of Korabl-Sputnik 5, the cosmonaut team, which consisted of 20 men, experienced its first fatality. Cosmonaut candidate Valentin Bondarenko was killed in a fire during a training exercise in an oxygen-rich isolation chamber.[4] It's not clear whether other cosmonauts were told of his death; the media didn't learn of Bondarenko's death - or even of his existence - until many years later, in 1986.[6]
